The Genome Analysis Core is a part of the Molecular Evolution Core.
The Genome Analysis Core focuses on several RT-PCR platforms plus single-cell encapsulation for RNA Seq 3' gene expression, 5' gene expression, and ATAC library production. Our technologies include:
The Fluidigm BioMark Genetic Analysis Platform is a system that is a fully integrated system enabling analysis of gene expression, genotyping, mutant detection, and absolute quantization of nucleic-acid sequences utilizing Dynamic Array Integrated Fluidic Chip (IFC) technology, which provides high-throughput real-time qPCR.
The Bio-Rad QX200 Droplet Digital PCR (ddPCR) provides absolute quantification of target DNA or RNA molecules for EvaGreen or probe-based digital PCR applications by using PCR-positive and PCR-negative droplets.
The Applied Biosystems StepOnePlus RT-PCR is designed to deliver precise, quantitative Real-Time PCR results for a variety of genomic research applications.
The BioRad ddSEQ™ Single-Cell Isolator in collaboration with Illumina can provide 3’ mRNA single-cell RNA Seq libraries by utilizing disposable microfluidic cartridges to co-encapsulate single cells and barcodes into subnanoliter droplets, where cell lysis, barcoding, and 3’ RNA Seq libraries can occur.
This 10X Genomics Chromium Single Cell system enables single-cell transcriptional profiling of up 500–10,000 individual cells per sample. 10X uses GemCode technology that samples a pool of ~ 750,000 barcodes to separately index each cell’s transcriptome. It does so by partitioning thousands of cells into nanoliter-scale GEMs, where all generated cDNA share a common 10x Barcode. Libraries are generated and sequenced from the cDNA and the 10x Barcodes are used to associate individual reads back to the individual partitions. The system allows for 3’, 5’, and ATAC library constructs.
The Lonza 4D Nucleofector System is an electroporation-based transfection method that enables the transfer of nucleic acids such as DNA and RNA into cells by applying a specific voltage and reagents.
Genome Analysis also provides other small services such as RNA quality control and spectrophotometer measurements for RNA, DNA, and protein (Nanodrop).
